Insert effects working one one song but not working on another song

If I apply effects inserts (eg X-Trem, Ampire, any effect) to one song in Studio One they work fine. However the same effects applied to another Studio One song don't work, ie the sound with the effect tued on is the same as the sound with the effect tued off. What goves whether effects work on a song - any ideas?
